摘要:
目的 探讨比索洛尔对培养的高血压大鼠(SHR)和Wistar大鼠心脏成纤维细胞(CFs)增殖及胶原合成的影响。方法 采用胰酶消化法培养CFs,采用3H胸腺嘧啶核苷(3HTdR)掺入法测定CFs的DNA合成功能,四氮唑蓝(MTT)比色法测定细胞增殖,3H脯氨酸(3HProline)掺入法测定胶原合成。结果 ①基础状态下,SHR组CFs的3HProline掺入量、3HTdR掺入量及MTT比色法A值均明显高于Wistar组。②不同浓度比索洛尔对两组大鼠CFs3HTdR掺入量及MTT比色法A值均无明显作用。③不同浓度比索洛尔以浓度依赖的方式抑制CFs 3HProline掺入量,与Wistar大鼠组相比,SHR组CFs受抑制的效应更强。结论 SHR的CFs细胞数目、DNA合成及胶原含量均存在异常,比索洛尔呈浓度依赖性抑制CFs的胶原合成,但对CFs增殖及DNA合成无明显作用。
Abstract:
AIM To evaluate the effects of bisoprolol on proliferation and collage synthesis of cultured rat cardiac fibroblast (CFs) derived from SHR and Wistar rat. METHODS CFs derived from 12weekold SHRs and Wistar rats were cultured with enzymatic dissociation. DNA synthesis was measured by,3HTdR incorporation. Cell numbers were determined by modified MTT array, 3Hproline incorporation was adopted to determine collagen synthesis. RESULTS ①Under basal condition,the incorporation of 3HProline and 3HTdR and A value in CFs derived from SHR were obviously greater than those of Wistar rats. ②Treatment with 0.01-10 μmol/L bisoprolol for 72 h did not produce significant differences of 3HTdR incorporation and A value in SHRs and wistar rats,compared with the control (P>0.05). ③ With the increase of concentration in the effects on bisoprolol, 3HProline incorporation in CFs of two groups decreased. CONCLUSION Bisoprolol inhibits collagen synthesis of CFs in a dosedependent manner. But it does not exert any effects on proliferation and DNA synthesis.
