摘要:
目的 探讨中药复方芪丹通脉片对大鼠缺血/再灌注损伤(RMMI)心肌细胞葡萄糖转运蛋白(GLUT4)的影响。方法 选用雄性SD大鼠36只,随机分为6组(n=6),即正常组、假手术组、单纯模型组、芪丹通脉片高剂量组和低剂量组、恬尔心组,各组均用生理盐水配制成等体积药液灌胃7 d,每日1次,采用冠脉结扎的方法建立大鼠心肌缺血/再灌注模型,将大鼠冠状动脉左前降支完全闭塞40 min,再灌注4 h后,速取再灌注区心肌组织,用免疫荧光、Western blot方法检测心肌细胞GLUT4的表达。结果 单纯模型组心肌细胞膜GLUT4蛋白明显表达,细胞中总的GLUT4含量较正常组增加;芪丹通脉片高剂量组和恬尔心组心肌细胞膜GLUT4蛋白表达明显增强,细胞中总的GLUT4含量较单纯模型组增加。结论 芪丹通脉片能明显增强RMMI大鼠心肌细胞总GLUT4的表达,并促进其向细胞膜转位,从而起到保护RMMI心肌的作用。
Abstract:
AIM To investigate the effects of Qidantongmai tablets on GLUT4 in the rat model of reperfusion mediated myocardial injury(RMMI). METHODS Thirtysix male SD rats were randomly divided into six experimental groups: control group, sham operation group, simple model group, QDTMT high dose group, QDTMT low dose group, and Diltiazem Hydrochloride group. Different drug or normal saline was administered to the animals at the same dosage once a day for seven days before operation. The model was established by ligating the coronary artery. After the levoanterior descending branch of coronary artery was occluded for 40 min followed by reperfusion for 4 hours, tissues in the reperfused region were sampled immediately and the protein levels of GLUT4 were determined by immunofluorescence and Western bolt. RESULTS In the simple model group, the expression of sarcolemmal GLUT4 was obvious and the level of total GLUT4 increased compared with that in the control group. In the QDTMT high dose group and the Diltiazem Hydrochloride group, the expression of sarcolemmal GLUT4 increased visibly and the level of total GLUT4 increased compared with that in the simple model group. CONCLUSION Qidantongmai Tablets have the effects to markedly promote the expression of GLUT4 in rats′ RMMI and to promote the translocation of GLUT4 to cell membrane, thus protecting the RMMI.
