摘要:
目的 探讨肿瘤坏死因子α(TNFα)mRNA在柯萨奇病毒B3 (Coxsackie virus B3,CVB3)病毒性心肌炎(viral myocarditis, VMC)小鼠心脏组织中的表情况,应用黄芪治疗后观察TNFα mRNA表达的变化。方法 4周龄纯种雄性BALB/c小鼠86只,随机分成4组。模型组40只,每只腹腔接种100 TCID50的以Hanks液稀释的CVB3m病毒液0.2 ml,分别于接种后1,3,7,9,11,21 d,随机取存活小鼠4只,将其处死,取心脏组织,冻存于-70℃冰箱中,待做RTPCR;黄芪用药组20只,同模型组接种病毒,0.5 h后开始以黄芪口服液给小鼠灌胃,每次0.2 ml, 2次/d,共10 d;未用药组20只,同模型组接种病毒,0.5 h后开始以生理盐水灌胃,每次0.2 ml, 2次/d,共10 d;正常对照组6只,每只腹腔接种Hanks液0.2 ml。于11 d处死用药组、未用药组及正常对照组组所有存活小鼠,取心脏组织冻存于-70℃冰箱中,待作RTPCR。采用半定量RTPCR检测心脏组织中TNFα mRNA的表达情况。结果 ①小鼠病毒性心肌炎心脏组织中TNFα mRNA于3 d表达开始增加,9 d达高峰,随后表达下降,21 d仍可见表达;②给予黄芪治疗可降低心脏组织TNFα mRNA表达水平(P<0.05)。结论 CVB3病毒性心肌炎小鼠心脏组织中TNFα mRNA早期即开始表达,并且其表达随时间而变化;黄芪可通过降低心脏组织中TNFα的表达而发挥其治疗VMC的有益的免疫调节作用。
Abstract:
AIM To investigate the effect of Astragalus Membranaceus (AM) on myocardial TNFα mRNA in mice with Coxsackie virus B3 viral myocarditis. METHODS Eightysix 4weekold male mice were randomly divided into 4 groups of A, B, C and D. Forty mice of group A were inoculated intraperitoneally with 0.2 ml CVB3 diluted in Hanks medium. At day 1, 3, 7, 9, 11 and 21 after virus inoculation, every 4 survived mice were killed, their heart tissues were taken out and stored at 70℃. Twenty mice of group B were perfused with AM to stomach after inoculated with CVB3 for 0.5 h, in a doze of 0.2ml and twice daily for 10 consecutive days. Twenty mice of group C were perfused with saline to stomach in the same dose as group B after inoculated with CVB3 for 0.5h. Six mice of group D as the control were inoculated intraperitoneally only with 0.2 ml Hanks medium. At day 11, all the survived mice were killed and their heart tissues were stored at -70℃. Semiquantitative RTPCR was then used to detect the expression of TNFα mRNA in the heart tissues. RERULTS ① TNFα mRNA increased 3 days after virus inoculation when compared to that of the normal control. The expression peaked at 9 days after inoculation, then decreased and could be seen 21 days after inoculation. ② Treatment with AM decreased the expression of myocardial TNFα mRNA when compared that treated with saline control (P<0.05). CONCLUSIONS The expression of TNFα mRNA increased in mice with CVB3 viral myocarditis and treatment with AM decreases myocardial TNFα mRNA expression.
