摘要:
目的 研究肾上腺髓质素(ADM)对血管紧张素II(Ang II)诱导的大鼠胸主动脉外膜成纤维细胞迁移的影响。方法 采用体外培养的大鼠胸主动脉外膜成纤维细胞,运用Transwell技术测定不同浓度ADM对外膜成纤维细胞迁移的影响,用RTPCR及Western blotting分析Ang II (1×10-6 mol/L)及不同浓度ADM干预后大鼠胸主动脉外膜成纤维细胞内骨桥蛋白的表达。结果 在Ang II (1×10-6 mol/L)趋化作用下,外膜成纤维细胞迁移活性较对照组显著增强,ADM可抑制Ang II刺激的细胞迁移,迁移细胞数目在一定范围内随着ADM浓度增加而减少;Ang II (1×10-6 mol/L)诱导骨桥蛋白呈高表达, ADM可下调这种表达,呈一定剂量依赖性。结论 ADM明显抑制Ang II刺激的外膜成纤维细胞迁移, 并且ADM可以抑制Ang II刺激的骨桥蛋白表达。
Abstract:
AIM To explore the effect of Adrenomedullin on angiotensin IIinduced migration in cultured rat vascular adventitial fibroblasts. METHODS Rat vascular adventitial fibroblasts were cultured in vitro and the effect of Adrenomedullin on the migration of adventitial fibroblasts was evaluated by transwell. The expression of osteopontin in adventitial fibroblasts stimulated in different conditions was measured by RTPCR and Western blotting. RESULTS Ang II stimulated adventitial fibroblasts migration and Adrenomedullin clearly inhibited Ang IIinduced migration of adventitial fibroblasts in a dosedependent manner between 1×10-9 and 1×10-7 mol/L. Ang II stimulated osteopontin expression in these cells and the inhibition by Adrenomedullin of Ang IIinduced adventitial fibroblasts migration was paralleled by a decrease in the expression of osteopontin. CONCLUSION ADM can inhibit adventitial fibroblasts migration and osteopontin expression induced by Ang II.
