摘要:
目的 探索成年糖尿病大鼠心室肌细胞成功分离的方法。方法 取体质量150~200 g的Sprague-Dawley大鼠20只,单次腹腔注射链脲佐菌素(STZ)建立DM大鼠模型,3周后取大鼠心脏行主动脉逆行灌流分离心室肌细胞;并在倒置显微镜下观察分离的单个心室肌细胞状态。结果 建模成功率达90%,糖尿病大鼠的血糖水平稳定维持在20 mmol/L以上;分离的单个心室肌细胞存活率达70%~90%,且纹理清晰状态好。结论 该糖尿病心室肌细胞的分离方法简单有效。
Abstract:
AIM To explore an effective method for successful isolation of ventricular myocytes of adult diabetic rats. METHODS The diabetic rat model was established by a single injection of streptozocin (STZ) peritoneally in Sprague-Dawley rats weighing 150-200 g. Ventricular myocytes were isolated by perfusion of enzymatic solution. The isolated ventricular myocytes were then observed under inverted microscope. RESULTS The successful establishment rate of diabetic rats was 90%, the blood glucose level stayed high above 20 mmol/L. The isolated ventricular myocytes were alive up to 70%-90% with good and clear stripes. CONCLUSION This simple method is an effective way for successful isolation of diabetic rat ventricular myocytes.
