摘要:
目的 探索基于数字视频分析的检测离体贴壁培养心肌细胞收缩特征参数的高效方法。方法 针对在二维平面上贴壁生长的心肌细胞自主收缩时每个特征位点实际上是在一维有限的空间上做往复运动的特点,本研究采用一维线性空间上的相关匹配算法替代以往二维块匹配跟踪算法,动态获取心肌细胞表面目标位点的运动位移矢量,以及速度、加速度和频率。结果 通过对合成的心肌细胞收缩标准视频测量表明该方法的收缩幅度测量结果与标准值一致,差异无统计学意义,并且能够跟踪实际视频中心肌细胞的收缩运动,动态获取收缩幅度、速度和加速度收缩特征参数。结论 该方法能够比较理想地动态检测贴壁生长的离体心肌细胞收缩的生物力学特征参数。
Abstract:
AIM To explore an effective approach based on digital video analysis to determine contractile characteristic parameters in adherent cardiac myocytes. METHODSIn accordance with the property that each specific point on the surface of adherent growth cardiac myocytes on a two-dimensional dish panel keeps moving actually in a periodic back-and-forth manner along a limited one-dimensional linear segment, we employed a correlation algorithm with one-dimensional linear searching region simplified from a typical block matching algorithm to determine the displacement vector, the contraction speed, the acceleration and the frequency. RESULTSFor synthetic standard video image sequence-containing contracting myoctes, the determined contractile amplitude values by this method exhibited close agreement with standard value without difference, showing no statistical significance. The dynamic contractile characteristic parameters including amplitude, velocity and acceleration could be obtained by automated tracking in practical videos. CONCLUSIONThis is a convenient method for quantifying biomechanical characteristic parameters of isolated adherent growth cardiac myocytes.
