摘要:
目的 探索磺脲类药物受体(SUR)亚型SUR1和suR2在仔鼠及成年正常大鼠心肌中的定位、分布及表达特征。方法 以 H-优降糖为放射性配体,运用体外受体放射自显影技术(In vitro receptor autoradiography)检测大鼠心肌中SUR1和SUR2的定位与分布模式,并用图像分析仪对其表达密度做半定量分析.结果 ① 在仔鼠及正常成年大鼠心肌组织中,SUR1为均匀分布;② 在仔鼠及正常成年大鼠心肌组织中,SUR2分布不均匀,室间隔,乳头肌等处的SUR2表达数目明显高于右心室及心房;③ 仔鼠与正常成年大鼠相比,在大鼠发育过程中,左心室的SUR2表达数目发生了明显变化,在仔鼠心肌组织中,与右心室相似,而在成年鼠,SUR2表达效目与室间隔相似。结论 SUR1和SUR2在心肌组织中的分布规律不同,SUR1在心肌各部位均匀分布,SUR2主要分布于心肌中的左室周边。
Abstract:
AIM To investigate the characteristics of expression and regional distribution of 2 subtypes of sulfonylurea receptors(SURl and SUR2)in the heart of immature and adult normal rat.METHODS With H-glibenclimade as radioligand,using in vitro receptor autoradiography,patterns of SUR expression and regional distribution were detected in tissue sections of immature and adult normal rat heart.Additionally,the expression density was analyzed semi-quantitatively by using an image-analyzing system.RESULTS Patterns of SUR1 and SUR2 expression and regional distribution as follows:The immature and adult normal rat heart demonstrated a homogeneous distribution for SUR1 and a heterogeneous distribution for SUR2,Additionally,patterns of distribution of SUR2 in immature and adult normal rat heart,especially in the region of left ventricule,were different during the postnatal development.C0NCLUSI0N The patterns of distribution of SUR1 and SUR2 in rat heart were different:SUR1 was homogeneous distributed while SUR2 was dominantly distributed in the left half in rat heart.
