摘要:
目的 U50488H 是阿片肽受体κ亚型的选择性激动剂, 本实验研究U50488H 预处理是否可增强心肌细胞对热应激的耐受性, 同时观察与热休克蛋白70 及90 表达的关系。方法 采用分离的大鼠心肌细胞作为研究对象,以心肌细胞释放乳酸脱氢酶(LDH) 的多少反映其损伤程度并用Westernblot 分析热休克蛋白表达的变化。结果 在5~ 30 Lmol/L 浓度范围内,U50488H 预处理30min 剂量依赖性的减低心肌细胞热应激后LDH 的释放。同时热休克蛋白70 和90 的表达明显增加。结论 U50488H 激活阿片受体后具有保护心肌细胞的作用, 其保护机制可能与调节热休克蛋白的表达有关。
Abstract:
AIM U50488H is the selective agonist of kappa opioid receptor. The present study was to investigate whether U50488H pretreatment increases rat ventricular myocytes to lerance against heat stress. METHODS Ventricular myocytes were isolated and pretreated with U50488H for 30 min, then they were subjected to heat stress at 44℃ for 30 min followed by 2 h culture at normal conditions (37℃). RESULTS The lactate dehydrogenase (LDH) release in the U50488H pretreatment group was decreased in the dose-dependent manner, and the expression of heat shock protein 70 and 90 were significantly increased. CONCLUSION Kappa opioid receptor activation with U50488H increases rat ventricular myocytes to lerance against heat stress with elevation of levels of HSP70 and HSP90.
