摘要:
目的 探讨肥大心肌胶原变化与左心室功能的关系。方法 采用大鼠腹主动脉缩窄模型, 术后10周检测左心室功能, 并用生化测定、病理检查结合计算机分析等方法检测左室心肌胶原形态、胶原容积分数(CVF)、血管周围胶原面积(PVCA)、胶原浓度(CC)、I ,III 型胶原比值(I/III)。结果 手术组左室压力下降最大速率(- dp/dtmax) 绝对值显著低于假手术组(P < 0.01) , 左室心肌CVF,PVCA,CC,I/III均显著高于假手术组(P<0.05) , 心肌胶原异常沉积。-dp/dtmax分别与CC 和I/III呈显著负相关( r1=-0.8904,r2=-0.8135,P<0.05)。Irbesartan治疗后,CVF,PVCA,CC,I/III可显著降低(P<0.05) , 异常沉积的胶原消失, 左室-dp/dtmax显著增高(P<0.01)。结论 压力超负荷左室重构早期舒张功能的降低主要与心肌胶原增生和胶原类型比值的改变密切相关。
Abstract:
AIM To investigate the relationship between change of myocardial collagen and left ventricular function. METHODS Using the methods of biochemistry assay and pathological examination combined with computed processing, the collagen volume fraction(CVF) ,perivascular circumferential area(PVCA),collagen concentration (CC),ratio of collagen type I totype III (I/III) were studied. Cardiac functional parameters were measured. RESULTS Left ventricular CVF,PVCA,CC,I/III, in aorta-constraction (AC) were significantly higher than those in sham-operated (SO) (P<0.05) , collagen abnormally accumulated, -dp/dtmax in AC was significantly lower than that in SO , -dp/dtmax was negatively carrelation with CC and I/III respectively (r1=-0.8904, r2=-0.8135,P<0.05). After treatment of Irbesartan, CVF,PVCA ,CC and I/III were reduced significantly (P<0.05) , Abnormally acculmulated collagen was disappeared. -dp/dtmax was obviouly increased (P<0.01). CONCLUSION Obvious hyperplasia of myocardial collagens and remodeling of collagen network occurrd after pressure overload, These changes induce left ventricular diastolic dysfunction.
