摘要:
目的 探讨无干预和血管升压素(AVP) 干预条件下, 心肌成纤维细胞(CFs) 的一氧化氮合酶—氧化氮(NOS-NO)系统活性的变化。方法 胰酶消化法分离、培养新生SD大鼠CFs, 采用硝酸还原酶法和分光光度法观察无干预和AVP干预条件下, 不同培养时间对CFs 的NOS-NO系统活性的影响。结果 ①无干预条件下, CFs 的NO 含量和NOS活性随培养时间延长而增高, 其中36h (42±5 Lmol·L-1, 37±5 U ·mL-1) 显著高于6 h (14±3 Lmol·L-1, 10±4 U·mL-1) 以及12h (21±3 Lmol·L-1, 15±3U·mL-1) (均P<0.05)。②AVP干预条件下, CFs的NO含量和NOS活性也随培养时间延长而增高, 其中24h(65±6 Lmol·L-1, 70±4U·mL-1) , 36h (62±1 Lmol·L-1, 69±6U·mL-1) 都显著高于6h (34±4Lmol·L-1, 36+2U·mL-1) 以及12h的(45±4Lmol·L-1, 45±1U·mL-1) (均P<0.05)。③AVP 干预条件下CFs 的NO含量和NOS活性均较无干预条件下显著增高, 且NO含量随NOS活性增高而增高, 二者呈显著正相关(无干预条件下r=0.837, P<0.01; AVP干预条件下r=0.936, P<0.01)。结论 AVP 提高CFs 的NOS-NO系统活性, CFs 的NOS-NO 系统活性与培养和AVP干预时间密切相关。
Abstract:
AIM To investigate the changes of nitric oxide synthase-nitric oxide (NOS-NO )system activity in neonatal rat cardiac fibroblasts (CFs) with and withou targinine vasopressin(AVP ). METHODS CFs were isolated by trypsin digestion method. Nitric acid reductase method and spectrophotometry were used to detect the NO contents and NOS activity respectively at different time points with or without AVP. RESULTS ①NO contents and NOS activity increased in a time-dependent manner without AVP (control group). NO contents and NOS activity of 36 h (42±uLmol.L-1, 37±5U.mL-1) were higher than those of 6h(14±3Lmol.L-1, 10±4U.mL-1) and those of 12h (21±3Lmol.L-1, 15±3U.mL-1 ) (both P<0.05). ②NO contents and NOS activity increased in a time-dependent manner with AVP (AVP group ).NO contents and NOS activity of 24h(65±6 Lmol.L-1, 70±4U.mL-1 ) and of 36 h (62±2Lmol.L-1, 69±6U.mL-1) were both higher than those of 6h (34±4 Lmol.L-1, 36±2U.mL-1) and those of 12h (45±4 Lmol.L-1, 45±1U.mL-1) (both P<0.05). ③NO contents and NOS activity of AVP group were both higher than those of control group. NO contents increased with the enhancement of NOS activity, and there was significant positive relevance between them (control group: r=0.837, P<0.01; AVP group: r=0.936, P<0.01).CONCLUSION AVP increases NOS-NO system activity in neonatal rat CFs. Whether with or without AVP, the NOS-NO system activity increases in a time2-dependent manner.
