目的 探讨新生动物同种主动脉带瓣管道(VAHC) 移植后免疫反应与钙含量之间关系。方法 体重150~ 200g 的新生新西兰兔24 只作为VAHC 的供体, 0. 9~ 1. 2 及2. 1~ 2. 9 kg 的幼兔和成年兔各12 只为受体, 建立新生兔的VAHC 移植模型。术后1 个月和6 个月分别取材作免疫荧光染色及钙含量测定。结果 免疫荧光检查显示各移植组VAHC 血管壁IgG 荧光染色结果均为阳性。半定量分析显示植入幼兔体内1 个月及成年兔1 个月和6 个月体内的阳性积分显著高于植入幼兔体内6 个月的VAHC (P < 0. 05)。各移植组VAHC 血管壁的钙含量及植入幼兔体内6 个月和成年兔体内瓣膜的钙含量显著高于对照组(P < 0. 05)。血管壁免疫荧光强度积分结果与钙含量呈明显的正相关( r= 0. 62, P < 0. 05)。结论 新生兔的VAHC 植入幼兔和成年兔体内后移植物的免疫反应程度与钙含量有明显的相关性。
Abstract:
AIM To investigate the crrelaion between immune respose and calcium content in implanted valved aortic homograft conduit (VAHC) of neonate rabbit. METHODS The animal model of VAHC was established via retroperitoneal pathway. Neonatal New Zealand rabbis(weight: 150~ 200 g, n= 24) were used as donors of the VAHCs, and young (0. 9~ 1. 2 kg, n=12) and adult rabbits (2. 1~ 2. 9 kg, n = 12) as recipients. The specimens were studied with immunofluorescence technique and their calcium contents were measured 1 month and 6 months after the implantation. RESULTS Immunofluorescence revealed the positive IgG fluorescent stain in VAHC walls of all the grafted groups. Semi-quantitative analysis showed that fluorescence stain in the VAHC walls of young recipients in 1 month and adult recipients after implantation was more intense than that in the VAHC walls of young recipients in 6 months after the implantation (P < 0. 05). The calcium content of VAHC walls in young recipients in 6 months and adult recipients was higher than that in the control group (P < 0. 05). The intensity of immunofluorescent stain and calcium content in the VAHC walls of all the grafted groups correlated obviously (R= 0. 62, P< 0. 05). CONCLUSION The intensity of immune response correlated well with the calcium content on implanted valved aortic homograft conduit (VAHC) of neonate rabbit.
