摘要:
目的:比较小鼠胚胎干细胞(ESC) 在不同诱导条件下分化为心肌细胞的分化比率。方法: 用鼠胚成纤维细胞(MEF) 作为饲养层促进ESC 增殖并抑制其分化, 用维甲酸(RA)、二甲基亚砜(DMSO)、转化生长因子B1 (TGF-B1)、激活素-A(activin-A) 为分化诱导剂, 采用三步法诱导ESC分化为心肌样细胞, 并比较各组分化比率。结果: 各组实验用的各种诱导剂均能诱导ESC分化为心肌样细胞, 尤以TGF-B1(2 ng/ml)、activin-A (20 ng/ml)及20% 胎牛血清(FCS) 组成的分化培养基可以使其分化比率高达88% , 显著高于其它各组(P< 0.01)。结论: 以MEF饲养层培养, TGF2B1、act ivin-A 作分化诱导剂可作为一种比较简便、稳定、高效的ESC分化条件。
Abstract:
AIM: To compare the different methods of inducing mouse embryonic stem cells (ESC)in vitro to differentiate into cardiomyocytes. METHODS: Mouse embryonic fibroblast (MEF)feeders were used to promote the growth and to inhibit the differentiation of ESC. During the inducing process, RA , DMOS, TGF-B1 and activin-A were used as inducing reagents and constituted six kinds of differentiating media. Then a three-step method was used to induce the differentiation of ESC. RESULTS: ESC could be induced in vitro to differentiate into cardiomyocytes in all groups, and the group induced by using TGF-B1 and activin-A acquired the highest differentiating ratio. CONCLUSION: TGF-B1 and activin-A were valuable reagents in inducing ESC to differentiate into cardiomyocytes.
