摘要:
目的:观察苯那普利对肾衰大鼠心脏的保护作用。方法: 实验分为假手术组(对照组)、肾大部切除肾衰组(RF组)、肾大部切除肾衰加苯那普利治疗组(RF+ Ben组)。苯那普利治疗剂量为10 mg·kg-1·d-1, 对照组用生理盐水灌胃。在实验第12周观察心脏的病理等改变。结果: RF 组大鼠心脏室间隔和左心室质量以及心脏质量/体质量比值均显著大于对照组。RF组心肌细胞肥大, 心内膜下及冠状动脉周围及其附近的心肌间质Ⅰ型胶原、Ⅲ型胶原及纤维连接蛋白沉积显著增加, 导致心肌间质纤维化, 而RF+ Ben 组其心肌细胞肥大和心肌间质纤维化程度明显减轻。结论: 肾大部切除肾衰大鼠可导致心室重构, 苯那普利具有明显的降压及防治实验大鼠左心室肥厚、心肌间质纤维化的作用。
Abstract:
AIM: To investigate the cardioprotective effects of Benazepril on subtotal nephrectomy renal failure rats. METHODS: To establish the subtotal nephrectomy renal failure model (RF) ,and the experimental rats were treated with Benazepril (10mg·kg-1·d-1) (RF+Ben) or normal saline as a control (Sham). Up to 12 weeks, the heart changes of experimental rats were observed. RESULTS: The myocardial mass of the in terventricular septal and left ventricle, and the ratio of heart weight/ body weight in the RF group were more higher than those in the sham group and in the RF+ Ben group. Myocardiac myocytes hypertrophia. The levels of collagen type, type Ⅲ , and fibronectin in endocardium and surrounding intracardial coronary arteries adventitia of the RF group were significantly increased as compared to the sham group. The levels of type Ⅰ、Ⅲ collagen and fibronect in were markedly decreased in the RF + Ben group as compared to the RF groups. CONCLUSION: The subtotal nephrectomy renal failure rats model may cause the left ventricular hypertrophy (LVH). Benazepril can prevent the LVH and myocardiac interstitial fibrosis.
