摘要:
目的:探讨心肌缺血后血管生成和碱性成纤维生长因子(basic fibroblast growthf actor,bFGF)表达的关系。方法:24只犬建立急性心肌梗死(myocardial infarction,MI)模型后随机分成对照组(MI区注射生理盐水15 ml)和实验组(MI区注射bFGF 50 mg与生理盐水的混合液15 ml)。每组观察4个不同时间点(术后第1,3,10和17天),每个小组3只犬。免疫组织化学方法检测各组心肌细胞中bFGF的表达及微血管数量。结果:除第1天外各个时间点的微血管数量实验组比对照组显著增多(对照组和实验组分别为:第3天:92±12,147±12,第10天:125±12,183±14;第17天:125±15,224±20;单位:个/视野),心肌缺血区治疗组bFGF的表达增多(对照组和实验组分别为:第1天:15±2,35±7;第3天:34±4,64±6,第10天:55±9,79±4;第17天:21±5,37±13),结论:局部心肌内注射bFGF有促进MI区域毛细血管形成的作用;心肌缺血区bFGF的表达与MI区域毛细血管的密度密切相关。
Abstract:
AIM:To evaluate the correlation between angiogenesis and the expression of bFGF in infarcted myocardium. METHODS:Twenty-four mongrel dogs were randomized into control group and the experimental group. After acute myocardial infarction was made,a mixfure of 50 mg of bFGF and 15 ml of saline was injected into the infarcted myocardium and its border zone in the experimental group,whereas 15 ml saline alone was given to the control dogs. Every 3 dogs in each group was studied on the lst,the 3rd,the l0th and the 17th days after the operation espectively.Angiogenesis and the expressions of bFGF of the two grups were evaluated by immunohistochemical studies with VⅢ factor and the antibody of and bFGF.RESULTS.Microvessel density was higher in the experimental group than in the control group excepton the first day (the controlg roup and the experimental group,respectively:the 3rd day,92士12and 147士1;the l0th day, 125士12 and 183士14;the 17th day,125士15 and 224士20). bFGF expressed more in experimental group the control group and the experimental group,respectively:the 1st day,15士2and 35士7;the 3rd day, 34士4 and 64士6;the l0rd day, 55士9 and 79士4;the 17rd day, 21士5 and 37士13). CONCLUSION:Intramyocardial administration of basic fibroblast growth factor helps to promote the growth of microvessels in acute myocardial infarction. There is a close relationship between angiogenesis and expression of bFGF in infracted myocardium.
