摘要:
目的: 探讨血管内皮生长因子(VEGF) 对人单核细胞源巨噬细胞(HMDM) 基质金属蛋白酶(MMPs) 的表达及其活性的影响。方法: 分别应用RT-PCR 和Western blot检测MMPs (MMP-2 和MMP-9)基因和蛋白表达,Zymography活性分析检测MMPs活性。结果: VEGF可明显增加人单核细胞源巨噬细胞(HMDM)MMP-2、MMP-9mRNA及其蛋白表达和活性。结论:VEGF可刺激HMDMMMP-2、MMP-9mRNA及其蛋白的表达, 增强其活性, 因而有可能促进动脉粥样硬化斑块内基质降解, 进而引起粥样硬化斑块破裂。
Abstract:
AIM: To observe the effect of VEGF on expression and activitiy of Matrix Metalloproteinase (MMPs). METHODS: The gene and protein expression of MMPs (MMP-2 and MMP-9) were respectively detected by RT-PCR and Western blot. MMPs’ activity was detected by Zymography. RESULTS: The expressions and activities of MMP-2 and MMP-9 mRNA of human monocytes were increased significantly by VEGF. CONCLUSION : As VEGF can stimulate the expressions and activities of MMP-2 and MMP-9 of human monocytes, it may increase the breakdown of matrix in lesions of atherosclerosis and enhance the lesions’vulnerability.
