目的:研究血管钠肽(VNP) 对人桡动脉(human radial artery , HRA) 的舒张作用及其机制。方法:采用离体血管 灌流的方法,观察VNP 对内皮完整和去内皮HRA 的舒张作用,以及82Br2cGMP、NPR2A、B 选择性抑制剂HS214221、鸟 苷酸环化酶选择性抑制剂美蓝(methylene blue , MB) 和Ca2 + 激活K+ 通道(Ca2 +2activated K+ channel , KCa) 的选择性抑 制剂TEA 对这一过程的影响。结果:VNP(10 210~10 - 6 mol/ L) 可剂量依赖性地舒张HRA ,该作用无内皮依赖性。82 Br2cGMP(10 - 7~10 - 3mol/ L) 可模拟VNP 的血管舒张效应。HS214221 (2 × 10 - 5mol/ L) 或MB (10 - 5mol/ L) 完全阻断 VNP 舒张作用。TEA(10 - 3mol/ L) 减弱VNP 的舒血管作用。结论:VNP 对HRA 具有不依赖内皮的舒张作用,它主要 是通过作用于NPR2A、B ,即钠尿肽的鸟苷酸环化酶耦联受体,升高细胞内的cGMP 水平。KCa可能是其舒张作用的 重要效应分子。为应用VNP 防治血管移植术后的痉挛提供了重要的理论依据。
Abstract:
AIM: To investigate the vasorelaxing effect of vasonatrin peptide (VNP) on human radial artery (HRA) and to reveal its underlying mechanism. METHODS : The vasorelaxing effect of VNP on HRA with and without endothelium was observed by perfusion in vitro. Effects of 8-Br-cGMP , HS-142-1 , the selective inhibitor of NPR-A and B , methylene blue (MB) , the selective inhibitor of GC , and tetraethylammonium (TEA) , the selective inhibitor of Ca2 + -activated K+ channel (KCa) , were also investigated. RESULTS : It was found that VNP from 10 - 10to 10 - 6 mol/ L induced a concentration2 dependent relaxation of HRA , which was independent of the endothelium. This effect was mimicked by 82Br2cGMP (10 - 7~10 - 3mol/ L) , and was abolished by either HS-142-1 at 2 ×10 - 5mol/ L , or MB at 10 - 5mol/ L. It was also attenuated by TEA at 10 - 3 mol/ L. CONCLUSION: The present study demonstrated that vasorelaxing effect of VNP on HRA is independent of endothelium , It results from an increase in the intracellular cGMP level by the activation of natriuretic peptide guanyl cyclase receptors in VSMCs , and KCa is an important mediator. The study has provided new and useful information for the use of VNP in the prevention and treatment of the spasm of transplanted blood vessels.
