目的 以自发性高血压大鼠 (SHR) 为研究对象,探讨血管紧张素转换酶抑制剂 (ACEI) 卡托普利长程治疗,对SHR左室肥厚(LVH)的改善及其可能机制。方法 20周龄雄性SHR 20只,随机分为卡托普利干预组(Cap组)和对照组(Con组),每组10只,观察卡托普利干预后13周内血压变化,并于观察期末测左心室质量/体质量(LVW/BW)及左心室组织cmyc基因的表达。结果 卡托普利 (100 mg·kg-1·d-1) 可使SHR的SBP显著降低,治疗4周后达最大降压效应,且该效应于第13周末仍可维持 (P<0.01);Cap组LVW/BW显著低于Con组(3.42±0.21 mg/g vs 3.96±0.18 mg/g,P<0.01),cmyc表达亦显著低于Con组(0.74±0.16 A vs 1.24±0.21 A,P<0.01)。结论 卡托普利长程治疗降压效应确切,并能显著改善SHR左心室肥厚,抑制原癌基因cmyc表达可能是其作用机制之一。
Abstract:
AIM To study the effects of long-term treatment of angiotensin converting enzyme inhibitor(ACEI) captopril on left ventricular hypertrophy(LVH) and oncogene cmyc expression in spontaneous hypertensive rats (SHR). METHODS 20 male SHRs aged 20 weeks were divided into two groups: 10 were given captopril, 100 mg·kg-1·d-1 by gavage for 4 weeks (Cap group) and another 10 were untreated group (Con group). Systolic blood pressure was measured each week. At the end of 13 weeks later, the rats were weighted and killed. Heart weight (HW), left ventricular weight (LVW) and LVW to body weight ratio (LVW/BW) were determined. The expression level of cmyc mRNA in left ventricular myocardium was measured by blot hybridization. RESULTS Compared with the Con group the systolic blood pressure decreased significantly in the Cap group, which reached the peak effective level in the 4th week 18.6±0.52 kPa vs 25.6±0.92 kPa,P<0.01, and maintained at this level till the 13th week 18.1±0.27 kPa vs 25.0±1.95 kPa, P<0.01. LVW/BW and cmyc expression in myocardium in Cap group was lowered significantly as compared with that of the Con group LVW/BW : 3.42±0.21 mg/g vs 3.96±0.18 mg/g,P<0.01; cmyc: 0.72±0.14 A vs 1.24±0.21 A,P<0.01. CONCLUSION Long-term treatment of captopril can definitely decrease blood pressure and inhibit LVH in SHR; the inhibition of oncogene cmyc expression in myocardium might play a role in its reversion of LVH.
